หลักการของ HPLC
HPLC เป็นเครื่องมือที่ใช้สำหรับแยกสารที่สนใจซึ่งละลายอยู่ในสารละลายผสม กระบวนการแยกสารจะเกิดขึ้นระหว่างเฟส 2 เฟส คือ เฟสอยู่กับที่ (column) กับเฟสเคลื่อนที่ (mobile phase) โดยสารจะถูกแยกออกมาในเวลาที่ต่างๆกัน ทั้งนี้ขึ้นอยู่กับความสามารถในการเข้ากันได้ของสารนั้นกับเฟสเคลื่อนที่หรือเฟสอยู่กับที่ สารตัวไหนที่สามารถเข้ากันได้ดีกับเฟสที่เคลื่อนที่สารนั้นก็จะถูกแยกออกมาก่อน ส่วนสารที่เข้ากันได้ไม่ดีกับเฟสที่เคลื่อนที่ หรือเข้ากันได้ดีกับเฟสอยู่กับที่ ก็จะถูกแยกออกมาทีหลัง และสารที่ถูกแยกออกมาได้นั้นจะถูกตรวจวัดสัญญาณด้วยตัวตรวจวัดชนิดต่างๆ แล้วจะแปรผลออกมาในรูปแบบกราฟที่เรียกว่า โครมาโตแกรม (chromatogram)

GC-MS ป็นเทคนิคหนึ่งที่เริ่มนิยมนำมาใช้กันอย่างแพร่หลาย เนื่องจากเป็นวิธีที่สามารถทำนายชนิดขององค์ประกอบที่มีอยู่ในสารได้อย่างค่อน ข้างแม่นยำโดยอาศัยการเปรียบเทียบ fingerprint ของเลขมวล(mass number) ของสาร ตัวอย่างนั้น ๆ กับข้อมูลที่มีอยู่ นอกจากนี้เทคนิคนี้ยังมีความสามารถในการวิเคราะห์ได้ทั้งใน เชิงปริมาณ (quantitative analysis) และ เชิงคุณภาพ (qualitative analysis) ได้อย่าง ถูกต้อง
